I just moved my dryer from one end of my basement to another and I had to make another outside opening for the vent. I used a 4.25" hole saw and this worked well. I made the opening over a basement window who's top is flush with the top of my foundation. I had to go through 2 2x8 and the sheathing. After, I thought that putting the dryer vent over the window was perhaps a mistake as I weakened the 2x8's that span the gap across the top of the window and foundation. Should I be concerned?

Yes, you should. I would re-enforce that rim joist with angle iron.
