I started to build cupola for my 8X14' barn style shed. I have plywood box and I need to build the roof. It comes from all four sides to a point in the middle. Should. they be cut on an angle of 45 degrees, going up the slope? Should it be made of plywood? Should I install 2x4 studs to support and nail, or is there another means of nailing? How would you then fasten to the roof? Is it different if it was not cut through the roof as a vent, or just left for decoration?

All you should need is a single spine under each of the four joints and I'd probably cut them at 45-degrees. The sheathing will be 7/16" OSB. As far as making the joins meet, I wouldn't bother because the shingles will cover them anyway. I certainly would not waste the ventilating opportunity and the easiest way to do that is to make the walls out of louvered doors. They're available in a variety of sizes and thicknesses.
